ALBA Time-Resolved Confocal Microscope

Confocal Microscope for multi-color confocal imaging, fluorescence lifetime imaging (FLIM) and fluorescence fluctuation spectroscopy (FFS)

User Manual available here.

Applications include:

  • fluorescence confocal imaging
  • fluorescence lifetime (FLIM) imaging
  • FRET and time-resolved FRET imaging
  • raster imaging correlation spectroscopy (RICS)
  • fluorescence correlation spectroscopy (FCS)
  • fluorescence cross-correlation spectroscopy (FCCS)
  • photon counting histogram (PCH)

Components:

  • ISS Alba time-resolved laser-scanning confocal system
  • IX-81 microscope body (Olympus), with objectives: U-Plan S-APO 60X water (UPLSAPO60XW), 1.2 NA, 0.28 mm working distance; UPlan FL 10X phase (UPLFLN10X2PH), 0.3 NA, 10 mm WD; LWD UPlan FL 20X phase (LUCPLFLN20XPH), 0.45 NA, 6.4-7.6 mm WD with collar
  • SPC-830 time-correlated single photon counting (TCSPC) board (Becker & Hickl) with 8 ps time resolution, up to 8 MHz count rate and 1024 x 1024 pixels resolution
  • SC-400-6-PP supercontinuum laser (Fianium) with 6W integrated average power, 5 ps pulse width (~100ps at fiber output), 1-80 MHz repetition rate, 400-2500nm wavelength range
  • Laser launcher with an IR cutoff at 750 nm and multiple wavelength (405, 488, 532, 561, 635 nm) output into one single-mode fiber
  • Two cooled, low noise avalanche photodiodes (APD) and two photomultiplier tubes (PMT), as well as a CMOS camera for brightfield imaging
